I have an OS plan with the 100 metres squares marked.
I want the ruler to coincide with the known 100m length
Then I can mark on layers various lines of say 7m or 25m long.
How can I set the ruler and guides to enable me to draw lines such as this.

If I understand you correctly, I'd load the map onto a layer and then adjust the scale to fit the ruler, so that 100 on the ruler was the same size as the 100 metre square.
I'd then draw my lines on another layer on top and keep the layer with the map on locked.

Eggs video is very good. My suggestion will work, but Eggs will put everything into the right units, so you don't have to worry about thinking too much.
