There are many tutorials which cover this type of work do a google search on this like most folk would do and then come back to the forum if and when you hit trouble or problems.

I'm about 50/50 these days between pen inking and digitally inking - some things [eg strips] I think I will always ink with a pen. I struggle to get the 'feel' digitally with a lot of stuff

having had a chance to look at the 'Turn Photos of People into Line Art' tutorial - its using pen tool to make paths and then stroking the brush [that you have previously defined] to the path

Jessy - this is an illustration of the way to do it if you have to with a mouse, but way clunky and fiddly compared to using a tablet and pen in my opinion, however it depends on the 'feel' you are aiming for I guess...

You might look at Akvis Coloriage. Not one that I have used, but you can try it for a coouple of weeks.
