Don't feel bad. I couldn't get it to work either no matter how I adjusted the Threshold slider....and it works in Painter 7!
Make sure your lines make closed areas so no color leaks out where lines are broken, then just use the Paint Bucket tool and click in the enclosed areas. If the Threshold is set right, it fills pretty well, right up to the edges of the black lines.
